Background: To explore the molecular mechanism of biological effects and potential long non-coding RNA (lncRNA) gastric carcinoma proliferation increases transcript 1 (GHET1) in acute myeloid leukemia (AML). Methods: Fluorescence in situ hybridization performed to determine the location GHET1. Quantitative polymerase chain reaction (qPCR) was performed to verify the RNA expression. GHET1 excess and knockdown achieved by transfection of the expression vector or short hairpin RNA. Western blotting, qPCR, Cell Counting Kit-8 assay, JC-1 staining and flow cytometry was conducted to measure the function GHET1. Dual luciferase reporter assay was conducted to confirm the relationship between microRNA 105 (miR-105) and Ras-related protein Rap-2B (RAP2B). Results: GHET1 localized in the nucleus of the cell line NB4. Expression GHET1 appointed in AML cell lines compared with normal bone marrow mononuclear cells. GHET1 knockdown led to inhibition of proliferation and promoted differentiation and apoptosis of AML cell lines. Subsequently, directly tied GHET1 miR-105 and miR-105 downregulated expression. MiR-105 suppressed excessive proliferation and induction of differentiation and apoptosis of AML cell lines. Additionally, RAP2B confirmed to be the target of miR-105 gene and demonstrated an inverse correlation between the level of their expression in AML cell lines; when miR-105 increased, the rate of Rap-2B decreases and vice versa. Conclusion: This study shows that GHET1 / miR-105 / Rap2B axis may be important signaling pathways involved in the development of AML.

Causal Relationship between Plasma Adiponectin and Body Mass Index: One-and two-sample two-way Mendelian Randomization Analysis on 460 397 Individuals

Methods: Information about 108 896 people from the Copenhagen General Population Study used in the Mendelian randomization analysis sample observational and a two-way, using the five genetic variants for BMI and 3 for adiponectin. For independent confirmation, information about 322 154 people from the GIANT consortium, and 29 347 people from the consortium ADIPOGen Mendelian randomization used in the analysis of two samples in two directions, using 68 genetic variants for BMI and 14 for adiponectin.
Results: In the observational analysis, an increase of 1 kg / m2 in BMI was associated with a 0.44 ug / mL (95% confidence interval: -0.46, -0.42) in plasma adiponectin, while an increase of 1 mg / mL in adiponectin plasma is associated with -0.11 kg / m2 (-0.12, -0.11) in BMI. In the causal genetic analysis, no relationship was observed between BMI and plasma adiponectin and vice versa. This kinase is activated by various environmental stresses and proinflammatory cytokines. p38 is expressed in the brain, heart, placenta, pancreas and skeletal muscle.
