Oxidized low-density lipoprotein (oxLDLs) and oxysterols play a key role in the development of endothelial dysfunction and atherosclerosis. Loss of function of vascular endothelium negative impact vasomotion, cell growth, adhesiveness and barrier function. While for some of these disorders plausible explanation can be provided from a mechanistic standpoint, for many others, the molecular players involved are not known. Caveolae, specific plasma membrane domains, has recently emerged as a target and mediator of oxLDL-induced endothelial dysfunction (ED).
Caveolae and their protein Caveolin-1 related (Cav-1) involved in oxLDLs / LDL transcytosis, especially through the scavenger receptor class B type 1 (SR-B1). Instead, oxLDLs LDL receptor endocytosis mediated by lectin-like oxidized 1 (LOX-1), whose activities depend on intact caveolae system. In addition, LOX-1 regulates expression of Cav-1 and vice versa.
On the other hand, oxLDLs may affect the plasma membrane cholesterol content / distribution thus affecting the architecture of caveolae, Cav-1 localization and related signals. Overall, this body of evidence shows that caveolae have both active and passive role in oxLDL-induced EC dysfunction. First as a mediator of lipid uptake and transfer in the subendothelial space, and then, as the target of a change in the composition / plasma membrane lipid dynamics resulting from increased levels of circulating oxLDLs.
Getting a better understanding of how oxLDLs interacts with endothelial cells (EC) and caveolae-mediated signaling pathways modulate, leading to ED, it is important to find new targets of intervention to address atherosclerosis and related clinical entities.
Thiol-based switching mechanism of stress-sensing companion
Thiol-based redox switches evolved as post-translational mechanisms that allow for efficient regulation of individual proteins to quickly respond to sudden environmental changes. While some of the functions of proteins need to be turned off to conserve resources and avoid potentially error-prone process, the protective function becomes important and need to be activated. In this review, we focus on the mechanism of thiol-based activation of the stress-sensing chaperones.
After the exposure to stress, the companion converted into a high-affinity binding platform for ongoing proteins and cells to protect against the accumulation of protein aggregates that are potentially toxic. their chaperone activity independent of ATP, a feature that is very important in conditions of oxidative stress, in which cellular ATP levels drop and canonical ATP-dependent chaperone no longer operate.
Likewise, reductive inactivation and release substrates require recovery of ATP levels, which ensures clients with foldases protein folding ATP-dependent. We will provide an overview on the different strategies that cells evolved to rapidly increase an ATP-independent chaperone on oxidative stress and provide mechanistic insight into how stress conditions used to convert the cellular protein overflow into the holding ATP-independent chaperones.
Both lysosomal integral membrane proteins form a complex MFSD1 and GLMP tight that confer protection from both interaction partners against lysosomal proteolysis. We are here enhanced molecular interactions of the two proteins and found that GLMP luminal domain, but not the transmembrane domain or short cytosolic tail, the inherent protection and mediate interactions with MFSD1.Our data support the finding that the interaction is important for the stabilization of the complex.
These results are supplemented by observations that N-glycosylation of GLMP in general, but not the type N-glycans (high-mannose-type or complex-type) or individual N-glycan chains, it is very important for protection. We observed that the interaction of the two proteins has already begun in the endoplasmic reticulum, and quantitative dependent on one another.
Both proteins can affect otherwise their intracellular trafficking to lysosomes in addition to protection from proteolysis. Finally, we provide evidence that can form homodimers MFSD1 both in vitro and in vivo. Our data repair complex interaction between intimate partner of the lysosomal transporter and accessory subunit.
TREND-DB-atlas transcriptome-wide dynamic landscape of alternative polyadenylation
alternative polyadenylation (APA) extend deep transcriptome complexity. Disruption of the APA can interfere with biological processes, ultimately resulting in disruption destroy. The main challenge in identifying the mechanisms and consequences of APA (and its disturbances) lies in the complexity of the RNA 3 ‘end processing, which involves poorly conserved RNA motif and multi-component complex consisting of more than 50 proteins.
It is more complicated that the RNA 3 ‘end maturation closely associated with transcription, RNA processing and even epigenetic (histone / DNA / RNA) modification. Here, we present TREND-DB , resource cataloging dynamic landscape APA after depletion> 170 proteins involved in various aspects of transcription, gene regulation co- and post-transcriptional, and epigenetic modifications further processing. TREND-DB visualization transcriptome dynamics 3 ‘diversification end (TREND) in a very interactive; APA provides global network maps and allows interrogating genes influenced by APA-specific regulator and vice versa. It also allows the analysis of functional enrichment of specific conditions WHAT affected genes, which show wide biological and clinical relevance in all conditions of RNAi.
