SARS-CoV-2 beta coronavirus is the driver COVID 19th etiology of the disease, which is mainly characterized by shortness of breath, persistent dry cough, and fever. Because they carry oxygen, red blood cells (RBCs) may play a role in the severity of hypoxemia in COVID-19 patients. The research combines state-of-the-art metabolomics, proteomics and lipidomics approach to investigate the impact of COVID-19 in the red blood cells of 23 healthy subjects and 29 diagnosed COVID molecular-19 patients. COVID red blood cells from 19 patients have elevated levels of glycolytic intermediates, accompanied by oxidation and fragmentation ankyrin, spectrin beta, and the N-terminal cytosolic domain band 3 (AE1). Significantly altered lipid metabolism was observed, in particular, short and medium-chain saturated fatty acids, acyl-carnitines, and sphingolipids. Nevertheless, there is no change in clinical hematology parameters, such as the RBC count, hematocrit or hemoglobin concentration of living cells on average, with only a small increase in the mean corpuscular volume. Taken together, these results suggest a significant impact of SARS-CoV infection-2 on the RBC membrane homeostasis structural protein and lipid levels. Increased RBC glycolytic metabolites consistent with the theoretical capacity increase hemoglobin to off-load oxygen as allosteric modulation function by high-energy phosphate compounds, perhaps to counteract hypoxia-induced COVID 19th. Conversely, because the N-terminus of AE1 stable deoxyhemoglobin and fine tunes the oxygen off-loading and rewiring metabolism towards hexose monophosphate shunt, the red blood cells of COVID 19 patients may be less able to respond to environmental variation in oxygen saturation of hemoglobin / oxidant stress while traveling from the lungs -paru to peripheral capillaries and vice versa.
MicroRNA-488 inhibits the proliferation and motility of tumor cells through downregulating FSCN1, modulated by Notch3 in breast carcinoma
As important modulators in several physiological processes, microRNAs (miRNAs) have been reported in various malignant tumors, including breast cancer. The current study investigated the function of new tumor suppressor microRNA, miR-488, and the molecular mechanisms of metastasis in breast cancer. CCK8 and transwell tests reveal that the regulated miR-488 significantly inhibited the rate of proliferation and migration of breast cancer cells. As a potential downstream genes, the level of mRNA and protein of FSCN1 oppressed by an increase in miR-488 and vice versa. luciferase assay showed that miR-488 directly bind to the 3’UTR of FSCN1 and pressed FSCN1 translation process. The promoter region of miR-488 directly bound by Notch3 and promoted the expression of miR-488 transcriptionally. The results of immunohistochemistry showed that in patients with breast cancer, the expression of Notch3 and negatively correlated with the level of significant FSCN1. Therefore, the current findings are predicted miR-488 molecules of tumor suppressor in breast cancer, and indicate that Notch3 / miR-488 / FSCN1 axis established and are involved in regulating metastasis of breast cancer, provide targeted therapies for patients with breast cancer.
Antarctic krill oil (AKO) is usually packaged by a protein material, increasing its oxidative stability. Protein shows great effect on lipid oxidation and induce co-protein-lipid oxidation. This study aims to determine the mechanism of co-oxidation AKO and whey protein (WP) or myofibrillar protein (MP) in the oil-in-water emulsion. Estimates of malondialdehyde (MDA) content, the molecular species of phospholipids, and pyrrole content resulting in increased and decreased levels of oxidation AKO (mainly phosphatidylethanolamine) by WP and MP, respectively.

Phosphorylation hCDC14A influenced by ZIPK regulate cell autophagy murine pancreatic islet β-TC3 on the stimulation of glucose
Purpose: Thinning of the island β cells play an important role in the onset of diabetes mellitus. Cell autophagy, as a self-healing process, contributing to maintaining metabolic homeostasis and may protect cells from apoptosis β small island in starvation or high glucose stress. However, the underlying regulatory networks autophagic processes in β cells of the island has not been fully explored.
Materials and Methods: Murine β-TC3 cells were treated with different concentrations of glucose, and the wild-type or mutant Ser484 human cell division cycle gene 14A (hCDC14A) was transfected. cell survival, proliferation and autophagy and secretion island studied. The mTOR and AMPK signaling pathway was investigated by Western blot. Zipper-interacting protein kinase was studied using mass spectrometry and immunoprecipitation.
Results: Expression of wild-type hCDC14A, but not Ser484 mutant hCDC14A, promoted cell survival, proliferation and autophagy is accompanied by increased secretion of the island and reduce cell apoptosis through inhibition of the mTOR pathway and activation of AMPK pathway in β-TC3 cells and vice versa. Furthermore, Zipper-interacting protein kinase (ZIPK), also known as DAPK3, was found to interact with hCDC14A especially for Ser484 phosphorylation, and ZIPK knockdown may affect phosphorylation hCDC14A and weaken the cell death or cell cycle modulation.
Conclusion: Overall, our results may provide new insights into the role of autophagy hCDC14A the island β cells and suggest potential therapeutic value of hCDC14A phosphorylation in the prevention and treatment of diabetes.
